The options for treating advanced melanoma are limited - regardless of whether the patient is a dog or a human. Because this deadly cancer is virtually resistant to chemotherapy and radiation in its late stages, new approaches are being investigated including vaccines that harness the immune system. For nine dogs that naturally developed canine malignant melanoma, treatment with a new DNA-based vaccine more than tripled their median survival from an expected 90 days to an average of 389 days.
The benefit of some cancer vaccines may be boosted by treating patients with an antibody that blocks a key protein on immune system T cells, according to a small, preliminary study led by researchers at Dana-Farber Cancer Institute and Brigham and Women's Hospital. The study tested the effect of a single injection of the antibody MDX-CTLA4 in nine patients who had previously been treated with cancer vaccines for either metastatic melanoma or metastatic ovarian cancer. The result, in every patient who had received a particular kind of vaccine, was widespread death of cancer cells and an increase in the number of immune system cells within the tumors ? evidence of a potent immune system attack.
A strong link exists between lifetime exposure to ultraviolet light, particularly lifetime sunburns, and the development of melanoma ? the most lethal form of skin cancer. Now, for the first time, scientists have identified a specific molecular pathway within cells that becomes mutated by ultraviolet light exposure, thereby speeding up melanoma development.
In a demonstration of vaccine therapy's potential for treating lung cancer, scientists report that a prototype vaccine boosted the natural immune response to tumors in a small group of patients with advanced non-small cell lung cancer (NSCLC). Researchers at Dana-Farber Cancer Institute have made a discovery that could help solve a mystery in cancer biology: how a sunburn acquired during a childhood day at the beach can develop into a deadly tumor decades later. The scientists report in the Feb. 4 Proceedings of the National Academy of Sciences that the sun's damaging ultraviolet (UV) rays target a series of biochemical signals inside the young skin cell, impairing the cell's ability to control its proliferation. Patients who develop melanoma on their face, head or neck can have the same early-diagnosis surgical procedure to see if their cancer might spread as patients whose cancer is on less delicate areas of the body, a new study finds. The report, from a team at the University of Michigan Comprehensive Cancer Center, opens the door for many more melanoma patients to benefit from a potentially life-saving technique called sentinel lymph node mapping. The results will be published in the Archives of Otolaryngology, a journal of the American Medical Association.
Scientists at the John Wayne Cancer Institute in Santa Monica, California have found a panel of molecular markers that could signal which patients might have the best results following vaccination for malignant melanoma. According to a study published in the January 15, 2003 issue of Cancer Research, a journal of the American Association for Cancer Research (AACR), the scientists showed that patient survival improved significantly if their tumors expressed higher levels for these markers, known as melanoma-associated antigens (MAAs).
Scientists at Emory University School of Medicine have identified a signaling pathway that is turned on when benign moles turn into early-stage malignant melanoma. Now, researchers at the University of Illinois at Chicago have isolated a protein secreted by bacteria that kills cancer cells but appears to have no harmful side effects. Tested in mice injected with human melanomas, the protein shrank the malignancies, but, in contrast with other studies using whole bacteria, caused no deaths or adverse reactions in the laboratory animals.
Treating precancerous breast cells with chemopreventive agents like tamoxifen limits the development of breast cancer in genetically predisposed women, according to a new study. Precancerous breast cells may also provide an effective mechanism for screening new and current chemopreventive agents, the study found.
