Discovery may lead to first medical treatment for Celiac disease

Results from a new study may lead to the first medical treatment for celiac disease, a hereditary digestive disease that can damage the small intestine and interfere with the absorption of nutrients from food. Celiac disease sufferers cannot tolerate gluten, a protein that is found in wheat, barley and rye. Celiac disease affects an estimated one in 250 Americans, mostly those of European descent, and there is no known medical treatment or cure.

Age, Lower Immunity Put Bypass Patients at Risk for Cognitive Impairment

Older patients with lowered immunity to certain common bacteria found in the gastrointestinal tract are more likely than younger patients to suffer cognitive decline after coronary artery bypass surgery, according to a new analysis by Duke University Medical Center researchers.
Within the gut resides a class of bacteria known as gram-negative bacteria. These bacteria can release endotoxins into the bloodstream as a result of action of the heart-lung machine -- which circulates the blood throughout the body while surgeons operate on a stopped heart -- triggering a cascade of immunological events including systemic inflammation.

FDA approves psoriasis drug

Alefacept, a specially designed molecule that blocks a specific immune-system reaction involved in the painful skin condition psoriasis, was approved for marketing today under the name Amevive. Biogen, Inc. of Cambridge, Mass., will market the drug. Alefacept traces its roots to research done at the U-M in the mid-1990s by a team led by former dermatology faculty member Kevin D. Cooper, M.D. The University and Biogen share the patent on the engineered molecule with Cooper, who is now chair of dermatology at Case Western Reserve University in Cleveland.

CRP improves cardiovascular risk prediction in metabolic syndrome

In women with metabolic syndrome, blood levels of C-reactive protein (CRP) can help predict cardiovascular risk, researchers reported today in Circulation: Journal of the American Heart Association. CRP is an indicator of inflammation. High levels of CRP have been linked to increased risk of heart attack, stroke, and type 2 (non-insulin dependent) diabetes.

New protein will help fight inflammation

A joint research project by scientists in Cardiff, UK, has developed a new protein, which could end the suffering of thousands. The research, by scientists at Cardiff University and the University of Wales College of Medicine (UWCM), and funded mainly by the Wellcome Trust, is designed to tackle the problem of chronic inflammation ? which can lead to serious disorders such as rheumatoid arthritis and bacterial peritonitis. Now a two-year development programme is getting under way to check the therapeutic potential of this new protein in the management of acute infection, to enable pharmaceutical and biotech companies to create a product for clinical use.

Scientists Discover How to Grow Cells That Suppress Immune Responses

Researchers have discovered how to grow a little-understood type of human immune cell. The cells, known as T-regulatory cells type 1 (Tr1), are thought to turn off unnecessary immune reactions and to block the action of immune cells that otherwise would attack the body and cause dangerous inflammation. The findings are reported in the Jan. 23 issue of the journal Nature.

Arthritis drugs may help the heart

Anti-inflammatory drugs used to treat arthritis may also benefit people with heart disease by improving blood vessel flexibility and reducing inflammation, according to a small study in today's rapid track report from Circulation: Journal of the American Heart Association. "Increasing evidence indicates that atherosclerosis is an inflammatory disease," says senior author Frank Ruschitzka, M.D., of the department of cardiology at University Hospital in Z?rich, Switzerland. "Thus, anti-inflammatory agents used to treat arthritis, such as COX-2 inhibitors, may not only reduce inflammation in the joints, but could possibly have that same anti-inflammatory benefit in the vessel wall. This study is the first to show that relationship."

Interleukin-6 May Lead to Drugs That Prevent Brain Injury From Diseases, Aging

A Mayo Clinic investigation of Interleukin-6, a hormone inside cells often considered a "bad actor" of the immune system because of its association with inflammation injuries and malignant diseases, shows that it also plays a therapeutic role in mice: it protects brain cells. Interleukin-6 -- called IL-6 for short by researchers -- may, in fact, be a "white knight" for mouse brain cells, or neurons, as brain cells also are called. These results, while early, may be promising for humans as well. The Mayo Clinic investigation is described in the Jan. 15 Journal of Neuroscience.

Protein in Eye May Help Fight Autoimmune Diseases

A protein found in the eye and involved in its "immune privilege" has prevented and halted autoimmune eye disease in animal models and promises to aid in preventing and treating other autoimmune diseases, including multiple sclerosis and diabetes, according to scientists at the Schepens Eye Research Institute at Harvard University. Immune privilege is a special property of the eye that allows the eye to protect itself without the inflammation caused by the body's conventional immune response to injury and infection.

Researchers ID protein behind tissue inflammation

Inflammation is usually a normal and beneficial response by the body to tissue injury or infection. But sometimes it can spiral out of control and lead to serious complications and diseases including pulmonary fibrosis, asthma and inflammatory bowel disease. Researchers have uncovered a mechanism that regulates the inflammatory response during tissue repair, providing the first specific molecular targets for developing ways to prevent highly destructive and potentially fatal inflammatory reactions.

Blood Proteins Put Dialysis Patients at Higher Risk of Heart Disease

A new study shows that two proteins are accurate predictors of heart attack or stroke in kidney dialysis patients. The research team found that high levels of C-reactive protein, a marker of inflammation, and low levels of albumin, a sign of malnutrition, had strong ties to heart disease in these patients, who are many times more likely to develop heart problems than the general population. "Both inflammation and malnutrition play an important role in the high risk of cardiovascular disease among dialysis patients," says Josef Coresh, M.D., Ph.D., lead author of the study and associate professor of epidemiology, medicine and biostatistics at Hopkins. "Testing for these proteins will allow us to sooner identify patients at high risk, and to manage heart disease risk factors more effectively."

Cholestrol Drug Could Lead to New Therapy for Multiple Sclerosis

While cautioning that their findings still must be evaluated in humans, University of California, San Francisco and Stanford University Medical Center researchers report that the cholesterol-lowering drug atorvastatin (Lipitor) significantly improved, prevented relapses or reversed paralysis in mice with an experimental disease that closely resembles multiple sclerosis. The study, reported in the November 7 issue of Nature, was conducted in mice with experimental autoimmune encephalomyelitis (EAE), the standard animal model for multiple sclerosis.

New hope for Alzheimer's vaccine

Researchers have discovered a way to refine an experimental Alzheimer's vaccine, a finding that could pave the way for new treatment and prevention of the debilitating disease that affects people's ability to think and recall information. Alzheimer's occurs when toxic biochemical compounds known as amyloid-beta peptides accumulate in the brain, forming plaque deposits and injuring nerve cells, which eventually causes dementia. In 2000, researchers at the Centre for Research in Neurodegenerative Diseases published a paper showing how the amyloid-beta peptide vaccine blocked the production of the plaques and reversed learning impairment. The vaccine stimulates the body's immune system into forming antibodies against the plaques in the brain, but it also caused inflamation. This new research claims to have found a way to better isolate the active epitope detected by antibodies. After testing a more refined, targeted amyloid-beta vaccine on mice, the scientists found that the antibodies generated by the vaccine cleared away the plaques -- improving cognitive function in the mice and leaving no evidence of brain inflammation.

Many neurological diseases occur when specific groups of neurons die because of nerve damage, toxins, inflammation, or other factors. A new study suggests that activity of a single gene can stop neurons from dying regardless of what triggers this process. The findings could lead to new ways of treating neurodegenerative diseases.

Valium-like drug helps treat lupus

A cousin to the anti-anxiety drug Valium has been shown in mice to reduce some of the symptoms associated with lupus. Lupus is an autoimmune condition in which the immune system attacks the body's own tissues. "The best available therapies for lupus haven't changed for many, many years," says U-M's Gary D. Glick, Ph.D., one of the lead authors on the study. "It's a disease where the mechanisms that normally prevent the immune system from attacking components of one's own body are defective. Because we do not yet understand what triggers lupus, it has been very difficult to develop lupus-specific therapies."
